Uranyless Acetone
UranyLess in Acetone is a reagent used for cryo-electron microscopy applications, particularly for freeze substitution techniques (AFS). This product serves as a safer alternative to traditional uranyl acetate, providing a solution for biological sample preparation while ensuring high-resolution imaging.

- Prepare the sample in an OsO₄ solution at -90°C for 10.5 hours.
- Increase the temperature for 0.5 hours.
- Immerse in an OsO₄ solution at -60°C for 10.5 hours.
- Wash with pure acetone at -30°C, then add UranyLess at -30°C for 8 hours.
- Gradually increase the temperature to 0°C to complete the protocol.
This product is designed to maximize image resolution while minimizing risks to the user, offering an alternative to uranium-based reagents for specimen preparation.
Protocol AFS : UranyLess Acetone version cryo for AFS application stain sample on bloc
- 8 hours 2% OsO4 solution at -90 °C
- 0.5-hour temperature increase
- 8 hours 2% OsO4 solution at -60 °C
- 0.5-hour temperature increase
- 8 hours 2% OsO4 solution at -30 °C
- Remove OsO4 solution and add pure acetone
- 1 hour washing
- Remove acetone and add UranyLess acetone version cryo for AFS application.
- 8 hours UranyLess acetone version at -30 °C
- 1.5 hours temperature increase
- 0 °C End of AFS
As soon as 0 °C was reached, the samples were washed three times with acetone and subsequently embedded in synthetic resin at room temperature.
With Uranyless Cryo:
